A custom scale range for a tile service does not work if Generation Method in the Generate Tile Cache Tiling Scheme tool is set to PREDEFINED.

In the Generate Tile Cache Tiling Scheme tool, select NEW from the Generation Method drop-down list, and type in the desired scales.
