In ArcGIS Online, labels and pop-ups set from the Visualization tab on the Item Details page are not reflected when viewed in the web map.

This is as designed. By default, any changes made to the properties of feature layers in the web map are stored in the map and remain disconnected from the source layer. This means that future changes to those properties in the source layer in the Visualization tab are not reflected in the map. Manage the layer properties in the web map to reset to the source layer properties if desired.
