Error

Changing the tile origin parameter when using arcpy.server.GenerateMapServerCacheTilingScheme does not actually change the tile origin.

Solución alternativa

Run the Generate Map Server Cache Tiling Scheme tool directly from ArcGIS Pro. Issue is only with arcpy.server.GenerateMapServerCacheTilingScheme.
