Additional Information

This is currently a limitation of ArcGIS Online-based feature services. Since case-insensitive queries are not supported, ArcGIS Pro cannot bypass this limitation. If ArcGIS Online implements a case-sensitive query capability, ArcGIS Pro will be able to use this functionality.

Workaround

Use the Export Features tool to work with a feature layer from the file geodatabase. Alternatively, in the attribute table, select features that only apply to the specific case wanted (lowercase or uppercase) after applying the initial definition query. Then, utilize the Show Selected Records mode in the attribute table.
