When creating a new enterprise geodatabase that is not registered as a data store and saving a map image as an offline service definition in ArcGIS Pro, the data seems to be somehow referenced after publishing the service definition file (.sd) in ArcGIS Server Manager.

This behavior is expected. Registering a database as a datastore only confirms that the server has access to the database, and that validation occurs only in ArcGIS Pro. The ArcGIS Server Manager site never runs any validation or analyzer. When the ArcGIS Pro publishing process is bypassed and instead creates an .sd file in ArcGIS Pro and then publishes it from the ArcGIS Server Manager site, the server assumes the publishing process is correct. It does not run any validation and tries to start the map or feature service by connecting to the database where the layers or tables are originally added in the map.
