In ArcGIS Online, thumbnails are stored in PNG format as described in https://doc.arcgis.com/en/arcgis-online/manage-data/configure-item-details.htm. PNG files support transparency. Thumbnails for portal items are stored independently from the item's metadata. When a file is added as a thumbnail to an item's metadata in ArcGIS Pro, it is converted to JPG format. JPG images do not support transparency. Therefore, the observed behavior is expected. Thumbnails in ArcGIS Pro are stored within the item's metadata XML document because there isn't an external repository in which to store independent files, as is possible in ArcGIS Online. The thumbnail format used by ArcGIS Pro cannot be changed for backward compatibility reasons.
