The 'RouteID' field drop-down menu in the 'Append Routes' tool does not recognize fields with a GUID data type.

Additional Information
A requirement for the route_id_field parameter is that the field type must match the RouteID field type of the target LRS Network. Refer to the following link for more information, https://pro.arcgis.com/en/pro-app/latest/tool-reference/location-referencing/append-routes.htm.Workaround
Create a new text field for RouteID, then run Calculate Field to copy the IDs into the text field. This text field is recognized in the RouteID drop-down, allowing users to proceed with the 'Append Routes' tool.
