The arcpy.SetRasterProperties tool is not able to set NoData to a null or empty value in a raster that contains a non-empty NoData value.

Workaround

arcpy.SetRasterProperties supports setting NoData to a value larger than the pixel depth. Setting NoData to 256 in an 8-bit raster appears to have the effect of making NoData unused, and does not update the raster to a larger pixel depth.
