The tool is behaving as designed. When using the ArcPy arcpy.management.CalculateField() function, a URL can be passed as the in_table. It is not necessary to use a Python API FeatureLayer object. Instead, use the calculate operation on the FeatureLayer object, as demonstrated in the documentaton: https://developers.arcgis.com/python/api-reference/arcgis.features.toc.html#featurelayerSteps to Reproduce
