In ArcGIS Pro, changing the grid unit labels to degree decimal minutes or degree minutes seconds does not reflect any changes in the layout.

This is as designed. The menu button controls the input options for the latitude and longitude boxes on the Element pane for the graticule. It does not control the label text. To change the label text (the values added to the layout), update the label formatting tags.
